Are all configurations possible: Yes
Are there license implication with some of the options: Yes
on-prem vs cloud the prereqs are the same for the base Linux machine configuration as far as size/disk/mem. Of course you need to make sure the machines can talk to each other but that it is the same as before.
Challenges will be speed. If the on-prem or EC2 node is closer to the data then you will see faster performance as the data has less to travel.
Now for license.
Option 1 and 2 nothing to talk about.
option 3 and 4. If you break out PwC and IDQ and install them on different domains (don't care where the domain lives) you CANNOT integrate the two products. meaning if installed on separate domains you cannot export a IDQ mapplet containing an address validation transform into PwC to cleanse address in your ETL load.
If you plan on never integrating the products then 3 and 4 are ok but if you want that integration 3 and 4 are not possible.

yes the picture is possible and many customers do it.
Ideally the Hybrid configuration is supported however ensure that you are not facing performance/stability issue just because of network latency between the nodes and the Domain DB.
Informatica mutli-node domains will be more stable when they are on same infrastructure(recommended) as it does not involve subnets, however if it is hybrid infrastructure you need to ensure network configurations are well tuned and there are no communication issues between the servers.
